چکیده

1. A new sensitive method for measuring &aminolevulinic acid (ALA) synthetase activity has been developed based on the incorporation of 14C-succinate into ALA. The method is extremely sensitive and will permit assay of the enzyme in normal tissues and in small amounts of tissue for which the existing calorimetric methods are inadequate. Difficulties due to aminoacetone synthesis are eliminated. 2. The procedure has been applied to liver homogenates and mitochondrial suspensions, to reticulocytes, and to bacterial extracts. 3. It has been found that the normal level of ALA synthetase activity in guinea pig liver mitochondria is even less than hitherto believed from calorimetric assays and cannot with confidence be said to be significantly above zero. 4. An inhibitor of ALA synthetase activity has been shown to be present in the insoluble fraction of normal liver mitochondria.

Bone marrow delta-aminolevulinic acid synthetase activity in hematological disorders.

An improved radiochemical method to measure the o-aminoJevulinic acid (ALA) synthetase activity in human bone marrow erythroblasts by incubating bone marrow cell homogenate with 14C-succinate and succinyl CoA generating system has been reported. Studies were made to obtain the optimal conditions for the enzyme assay and to confirm the reliability of the assay procedures. ALA-synthetase in eryth...

Heme synthetase deficiency in human protoporphyria. Demonstration of the defect in liver and cultured skin fibroblasts.

The final step in heme biosynthesis is chelation of porphyrin with Fe++ catalyzed by the mitochondrial enzyme heme synthetase. We have employed a sensitive radiochemical assay for this enzyme, using 59Fe and deuteroporphyrin or protoporphyrin as substrates. In this method iron is maintained in the ferrous state, oxygen is excluded from the incubation system, and labeled heme product is extracte...

Radiochemical Microassay of &AminolevulinicAcid Dehydratase Activity in Whole Blood and Bone Marrow

We describe an assay for 5-aminolevulinic acid dehydratase (EC4.2.1.24) activity. Radioactive [14Cjporphobilinogen, formed by action of this enzyme on [14CJo-aminolevulinic acid, is purified by passage through an ion-exchange chromatographic column before measurement with a liquid scintillation counter. The radioactive substance in the final solution was identified as solely [14C]porphobilinoge...

delta-Aminolevulinic acid synthetase in erythroblasts of patients with pyridoxine-responsive anemia. Hypercatabolism caused by the increased susceptibility to the controlling protease.

Properties of delta-aminolevulinic acid synthetase in erythroblasts of patients with pyridoxine-responsive anemia were investigated with special reference to the protease in mitochondria of erythroblasts. delta-Aminolevulinic acid synthetase activity in erythroblasts of patients with this disease before treatment was extremely decreased, whereas it gradually increased in parallel with the impro...

Purification of a specific activator of solubilized delta-aminolevulinic acid synthetase from rat liver mitochondria.

A protein capable of activating 6-aminolevulinic acid synthetase has been purified from rat liver mitochondria. By treatment of the mitochondrial inner membrane-matrix fraction with Lubrol a soluble and active preparation of 6-aminolevulinic acid synthetase has been obtained. Ammonium sulfate fractionation yielded a 33 to 40% saturated fraction containing catalytic 6-aminolewlinic acid syntheta...
